The primary detector for the CGSC is a BioTek Synergy-2 microplate reader.
Our reader supports three major modes of readout,
- Fluorescence
- Optical Absorbance
- Luminescence
Each of the modes is capable of multiple sub-modes to allow for the widest range of assays to be used.
Operating in fluorescence mode the reader can take four different types of measurements:
- Intensity
- Polarization
- FRET
- Time Resolved
A wide selection of filters and dichroics make most dyes and dye-pairs accessable to the reader.
Filters exist for: Tryptophan, DAPI, Fluorescein, GFP, EYFP, CY3, TRITC, Rhodamine, Texas Red, CY5.
At the moment polarization readings are limited to Fluorescein, GFP, CY3, Rhodamine, Texas Red, CY5.
If you would like to make use of another fluor, please e-mail or call.
We will likely be able to adjust our filters, or get a new filter as part of your experiment.
Absorbance and Optical Density measurements are made using a tunable monochromator and a dedicated
detector. Wavelengths from 280 nm through 1000 nm are accessible,
making pathlength corrections possible on a per-well basis.
Optical densities (OD) from 0.05 to 1.5 can be read with little or no distortion.
Highly sensitive luminescence measurements can be made using a specialized optical path. The reader is equipped with reagent injectors for flash luminescence readouts as well as glow based systems.
